SUMMARY

Fundamentals in Histochemistry and Optical Microscopy Applied to Studies in Plants
First Theoretical-Practical Course

Summary
With a series of short courses, we begin our training in various theoretical and technical aspects in the morpho-anatomy and ultrastructure of plants using traditional and contemporary methods and technologies in microscopy; this also includes plant histochemistry techniques related to light microscopy. We offer it to anyone interested in the field of Botany and related disciplines. Currently, plant anatomy is a discipline in high demand in the market, as both a basic science and a tool that facilitates the solving of various problems at the taxonomic, evolutionary, ecological, and biotechnological levels.
Knowledge of the external and internal structure of plants is vital to understanding and explaining specific processes unique to them, and their interrelationship with the environment. Taking into account the low number of publications with a focus on morpho-anatomy in Colombia, it is necessary to carry out more structured studies of plants in order to grow our database and enrich our knowledge of the flora in our country.
As institutions in charge of divulging this information to the field of botany and updating the community on new techniques and state-of-the-art equipment, we see the need to train professionals who can lead these long-term investigations, and, through the precise application according to the type of study to be carried out and the type of plant material to be processed, make use of the present-day technologies that are being integrated into our country.
General Objective
Train students and professionals in practices such as histochemistry and optical microscopy in relation to plant morpho-anatomy.
Specific Objectives
Train participants in, the basic concept and application of handling techniques for fresh plant material preparations, and , the learning and application of different staining methodologies and basic optical microscopy techniques.

Bachelor and postgraduate degrees in Biological Sciences (2002), Master (2005) and Doctor (2008) in Plant Biology from the State University of Campinas. At the time of the cusro, the professor of the Department of the Institute of Biosciences of the University of São Paulo Botany, works in the area of Plant Anatomy, with emphasis on secretory structures and floral development and evolution.

1 - Introduction to Scanning Electron Microscopy (SEM):
Features High vacuum AND low vacuum scanning electron microscope.
-Electronic optics: Electron source (tungsten filament and field emission).
-Electromagnetic: properties, aberrations. Resolution and depth of field. Magnification. Electron interaction with matter. Types of emission: secondary electrons, backscattered and characteristic X-rays.
